Risk of Genome-Wide Association Study-Identified Genetic Variants for Non-Hodgkin lymphoma in a Chinese Population
<正>Background Recent genome-wide association studies have identified 12 single nucleotide polymorphisms(SNP) associated with the Non-Hodgkin lymphoma subtypes in Caucasians.There are different incidence rate of NHL and different proportion of NHL subtypes in Chinese compared with those in Caucasians.This study evaluated the effects of these identified SNPs on susceptibility to NHL in a Chinese population.Methods We assessed the associations of these 12 SNPs with NHL in a case-control study that consisted of 810 cases and 1,543 controls.Odds ratios(OR) and 95%confidence intervals(CI) were computed by logistic regression,and cumulative effect of risk alleles were also calculated.Results We found that the allele frequencies of these SNPs in our study population are significantly different from those in Caucasian populations,with the 5 SNPs(rs17483466,rs757978,rs13397985,rs735665 and rs11083846) being extremely rare in Chinese.Two SNPs(rs872071 in the 3'-UTR of IRF4 and rs10484561 in the human leukocyte antigen(HLA) classⅡ) were significantly associated with NHL risk in our study population,with the OR of 1.20,(95%CI, 1.04-1.38;P_(trend)=0.011) for rs872071 and OR of 1.97 for rsl0484561(95%CI, 1.61-2.42;P_(trend)=1.05×10~(-10)) calculated using additive model.The associations of these two SNPs with the risk were seen in both T-cell NHL and B-cell NHL.With increasing number of risk genotypes,a cumulative effect was observed with the ORs being gradually elevated(P_(trend)=3.52×10~(-9)).Compared with carrying no risk genotype,individuals carrying 1 or 2 risk genotypes had an adjusted OR of 1.41 (95%CI,1.17-1.71;P=0.004) or 2.52(95%CI,1.85-3.44;P=5.98×10~(-9)), respectively.The other SNPs had no significant associations with NHL risk in the Chinese population.Conclusions These results suggest that the SNPs rs872071 and rs10484561 contribute to the risk of developing NHL in the Chinese population individually and jointly
