Foundation of protein preparation method and 2-DE analyses of anti-radiation proteins in Antarctic ice microalga Chlamydomonas sp.
<正>Protein preparation is the key step of two - dimensional gel electrophoresis. Antarctic ice microalga Chlamydomonas sp. was used as material in our experiments, from which protein extract methods were compared, and the results showed that improved Trichloroacetic acid (TCA) -acetone fractional precipitation method was one of the fittest methods to protein extraction from Antarctic ice algae. At the same time, we founded a complete 2 - D electrophoresis system fit to Antarctic ice alga proteins. We also analyzed the anti - radiation proteins in Chlamydomonas sp. by 2 - D gel electrophoresis, and two new peptides (21 and 23 kD) were found after exposing to UV radiation.
