Sequence analysis and functional characterization of the antifungal biosynthetic pathway from Burkholderia pyrrocinia strain Lyc2

  Bacterial strain Lyc2 was isolated from tobacco rhizosphere and showed strong antifungal activities.The 16 S rRNA and multilocus sequence typing(MLST)analyses revealed that strain Lyc2 belongs to Burkholderia pyrrocinia.Bioassay results indicated Lyc2 showed significant antifungal activities against a broad range of plant and animal fungal pathogens.Mutagenesis analysis revealed that a 55.2 kb gene cluster was essential for antifungal activities of strain Lyc2.In this study,the mutant Lyc2-MT11(orfll::Tn5) showed significant reduction in antifungal activity against Geotrichum candidum compared with that of the wild-type strain.Analysis of the protein sequence suggests that the or/11 gene encodes a flavin-dependent monooxygenase.HPLC was used to verify the production of antifungal compounds.Multiple antibiotic and secondary metabolized biosynthesis gene clusters predicated by antiSMASH revealed the broad spectrum of antimicrobials activities of the strain.Our results revealed the mechanisms of antifungal activities of strain Lyc2 and expand our knowledge about production of antifungal compounds in the bacteria Burkholderia pyrrocinia.……
