Mapping of the minimal epitopes for three coat protein specific monoclonal antibodies commonly used to detect Potato virus Y

  Potato virus Y(PVY)(genus Potyvirus)is the most economically damaging in Solanaceous plants including potato,tobacco and pepper.Enzyme-linked immunosorbent assay(ELISA)has been widely used for detection of PVY.Mab1 128,Mab1129 and Mab1130 were three of widely used commercial monoclonal antibodies to distinguish different PVY strains.In this study we mapped three minimal epitopes within coat protein for those three antibodies using peptide scanning,alanine replacement,and residue deletion.All mapped epitopes located within 30 N-terminal residues of coat protein.Sequences alignments showed that these epitopes are relatively conserved.Mutagenesis results of the most variable position within the epitopes indicated that some of those virus isolates could be escaped the detection.……
