p75 is mainly responsible for abeta toxicity but not its internalization

  Background:Accumulating evidences indicate that the intraneural beta-amyloid peptide(Aβ) aggregates is early than extraneuronal plaque formation and may be the pathological biomarker for the onset of Alzheimer s disease(AD).p75 is receptor of Aβand there is interaction between them. The purpose of this study is to certificate if p75 helps Aβinternalization to find new therapeutic target for AD. Results:p75 is required for Aβ1-42 oligma induced cell apoptosis but not necessary for Aβ1-42 oligma internalization in pc12 cells and primary neurons.Internalized Aβ1-42 oligma is mainly colocaolizated with beclin-1(biomarker of autophagysome) and the internalization process can be affected by cholesterol content in culture media. Conclusions:p75 may not mainly responsible for Aβinternalization but responsible for Aβinducing apoptosis.The membrane fluidity and its interaction with Aβ1-42 oligma might results in Aβinternalization.……
