Characterization and Expression of the Genes including CD63, p8 and cathepsinL/B involved in the immune response in amphioxus

  We report the characterization and expression of the genes including CD63, p8 and cathepsinL/B involved in the immune system in the amphioxus Branchiostoma be(?)cheri tsingtauense.The cDNA encoding CD63 was identified for the first time from the gut cDNA library of amphioxus. Primary structural examination showed that the protein encoded by the cDNA contained four potential transmembrane domains characteristic of TM4SF proteins and a conserved CCG motif in the putative major extracellular loop. RT-PCR analysis exhibited that AmphiCD63 mRNA was abundant in muscle, ovary, foregut including hepatic caecum and hindgut, while it was present at considerably lower levels in notochord and gill and absent in testis. The full - length Amphip8 cDNA consists of 1032 bp and encodes 79 amino acids. Analysis of the p8 primary structure suggested the presence of a motif of nuclear targeting, indicating that p8 may function within the nucleus. RT - PCR analysis showed that Amphip8 mRNA was abundant in muscle, notochord, ovary, gill, gill, gut including hindgut, while it was present at lower levels in foregut. Amphioxus cathepsin L includes conserved motifs ERFNIN, and the amphioxus cathepsin B contains a conserved occluding loop. RT - PCR, Northern blot and section in situ hybridization experiments showed that both AmphiCL and AmphiCB expressed in all tissues exmined, with the most abundance in the hepatic caecum and hind - gut, suggesting the role of food digestion for the two enzymes. Whole mount in situ hybridization analysis revealed that AmphiCL was firsdy detected in the primitive gut of 2 - day larvae, whereas AmphiCB was firstly detected in the primitive gut of 1 - day larvae, suggesting that AmphiCB plays its physiological role in advance compared with AmphiCL . AmphiCL expression was down regulated in the guts of amphioxus when fed with spirulina and up regulated when exposed to LPS. AmphiCL may play a role in inflammatory reaction in amphioxus.……
